A C-type lectin from Bothrops jararacussu venom can adhere to extracellular matrix proteins and induce the rolling of leukocytes J. Venom. Anim. Toxins incl. Trop. Dis.
Elífio-Esposito,S. L.; Hess,P. L.; Moreno,A. N.; Lopes-Ferreira,M.; Ricart,C. A. O.; Souza,M. V.; Hasselman-Zielinski,F.; Becker,J. A.; Pereira,L. F..
Purification of a lectin from Bothrops jararacussu venom (BjcuL) was carried out using agarose-D-galactose affinity gel. MALDI-TOF gave a major signal at m/z 32028, suggesting the presence of a dimmer composed of two identical subunits. Divalent cations were required for the lectin activity, as complete absence of such ions reduced hemagglutination. BjcuL was more effective at neutral pH and showed total loss of activity at pH values below 4.0 and above 9.0. Its agglutinating activity remained stable at 25°C until 60min, but increased when at 35°C for at least 15min. Adhesion assays to extracellular matrix (ECM) glycoproteins showed that the biotinylated lectin (0.039-5.0µg/100µl) was capable of binding to fibronectin and vitronectin in a dose-dependent...

Arachidonic acid triggers an oxidative burst in leukocytes BJMBR
Pompeia,C.; Cury-Boaventura,M.F.; Curi,R..
The change in cellular reducing potential, most likely reflecting an oxidative burst, was investigated in arachidonic acid- (AA) stimulated leukocytes. The cells studied included the human leukemia cell lines HL-60 (undifferentiated and differentiated into macrophage-like and polymorphonuclear-like cells), Jurkat and Raji, and thymocytes and macrophages from rat primary cultures. The oxidative burst was assessed by nitroblue tetrazolium reduction. AA increased the oxidative burst until an optimum AA concentration was reached and the burst decreased thereafter. In the leukemia cell lines, optimum concentration ranged from 200 to 400 µM (up to 16-fold), whereas in rat cells it varied from 10 to 20 µM. Initial rates of superoxide generation were high,...

Composto homeopático reduz a liberação de ânion superóxido pelas células mononucleares de ema (Rhea americana) Arq. Bras. Med. Vet. Zootec.
Bertoldo,W.R; França,J.L; Fernandes,L.T.O; França-Botelho,A.C.
The action of the "Stress Factor Ostrich (Arenales - Fauna and Flora)" was tested in the release of superoxide anion by cells in the peripheral blood of rhea (Rhea americana). Sixteen samples of 0.5mL of venous blood were collected through the jugular vein in the morning and placed in heparinized tubes. The leukocytes were separated into polymorphonuclear (PMN) and mononuclear (MN). The production of superoxide anion by phagocytes of peripheral blood was determined using the chromogen ferricytochrome C. There was a reduction of superoxide by MN cells in the presence of "Stress Factor Ostrich" indicating a positive influence of product against oxidative stress. Furthermore, future researches, such as the evaluation of other reactive oxygen intermediates and...

Contribution of CD4+ T cells to the early mechanisms of ischemia- reperfusion injury in a mouse model of acute renal failure BJMBR
Pinheiro,H.S.; Camara,N.O.S.; Noronha,I.L.; Maugeri,I.L.; Franco,M.F.; Medina,J.O.A.P.; Pacheco-Silva,A..
Renal ischemia-reperfusion (IR) injury is the major cause of acute renal failure in native and transplanted kidneys. Mononuclear leukocytes have been reported in renal tissue as part of the innate and adaptive responses triggered by IR. We investigated the participation of CD4+ T lymphocytes in the pathogenesis of renal IR injury. Male mice (C57BL/6, 8 to 12 weeks old) were submitted to 45 min of ischemia by renal pedicle clamping followed by reperfusion. We evaluated the role of CD4+ T cells using a monoclonal depleting antibody against CD4 (GK1.5, 50 µ, ip), and class II-major histocompatibility complex molecule knockout mice. Both CD4-depleted groups showed a marked improvement in renal function compared to the ischemic group, despite the fact that...

Disintegrins: integrin selective ligands which activate integrin-coupled signaling and modulate leukocyte functions BJMBR
Barja-Fidalgo,C.; Coelho,A.L.J.; Saldanha-Gama,R.; Helal-Neto,E.; Mariano-Oliveira,A.; Freitas,M.S. de.
Extracellular matrix proteins and cell adhesion receptors (integrins) play essential roles in the regulation of cell adhesion and migration. Interactions of integrins with the extracellular matrix proteins lead to phosphorylation of several intracellular proteins such as focal adhesion kinase, activating different signaling pathways responsible for the regulation of a variety of cell functions, including cytoskeleton mobilization. Once leukocytes are guided to sites of infection, inflammation, or antigen presentation, integrins can participate in the initiation, maintenance, or termination of the immune and inflammatory responses. The modulation of neutrophil activation through integrin-mediated pathways is important in the homeostatic control of the...

Effect of fatty acids on leukocyte function BJMBR
Pompéia,C.; Lopes,L.R.; Miyasaka,C.K.; Procópio,J.; Sannomiya,P.; Curi,R..
Fatty acids have various effects on immune and inflammatory responses, acting as intracellular and intercellular mediators. Polyunsaturated fatty acids (PUFAs) of the omega-3 family have overall suppressive effects, inhibiting lymphocyte proliferation, antibody and cytokine production, adhesion molecule expression, natural killer cell activity and triggering cell death. The omega-6 PUFAs have both inhibitory and stimulatory effects. The most studied of these is arachidonic acid that can be oxidized to eicosanoids, such as prostaglandins, leukotrienes and thromboxanes, all of which are potent mediators of inflammation. Nevertheless, it has been found that many of the effects of PUFA on immune and inflammatory responses are not dependent on eicosanoid...

Effect of PHA and LPS on the Activity of Nonspecific Cytotoxic Cells and Macrophages of Rainbow Trout (Oncorhynchus mykiss) Gayana
Gómez,Leonardo A; Cortés,Raúl; Smith,Carlos T.
This work studied the effect of the lectin phytohaemagglutinin (PHA) and bacterial lipopolysaccharide (LPS) on the activity of nonspecific cytotoxic cells (NCCs) and macrophages of peripheral blood in the rainbow trout. NCCs are cells of the innate immunity, homologues to Natural Killer (NK) cells of mammals, capable of eliminating cells infected with intracellular pathogens and also tumoral cells. Their lytic activity is mediated by the perforin/granzyme system and the Fas receptor-Fas ligand (FasR-FasL) system. Activity of NCCs was evaluated by their capacity to lyse HL-60 tumor cells. On the other hand, macrophages eliminate microorganisms by several mechanisms, including the production of reactive oxygen species (ROS) and nitric oxide (NO). Results...

Machado,Michel M.; Oliveira,Luis F.S. de; Zuravski,Luisa; Souza,Raul O. de; Fischer,Paula; Duarte,Jonathaline A.; Rocha,Manoelly O.; Güez,Camila M.; Boligon,Aline A.; Athayde,Margareth L..
ABSTRACT Euphorbia tirucalli (L.), commonly known as aveloz, has been indiscriminately used in popular medicine to treat various illnesses. However, some components can have devastating consequences. Injury to a cell's genetic material can cause mutations, cancer, and cell death. Our main goal in this work was to evaluate the genotoxic and cytotoxic effects of E. tirucalli extract on human leukocytes. For this purpose, we performed a phytochemical analysis to evaluate the plant's components. In the second step, we treated cultured human leukocytes with different concentrations of the dry extract of the plant and then evaluated the oxidative and genotoxic profiles of these leukocytes. We found that at 1% and 10% concentrations, the aveloz extract acted as a...

Mendonça,V.A.; Alvim de Melo,G.E.B.; Araújo,M.G.; Borges,V.O.; Costa,R.D.; Martins-Filho,O.A.; Teixeira- Carvalho,A.; Sathler-Avelar,R.; Teixeira,M.M.; Teixeira,A.L..
Leprosy is caused by Mycobacterium leprae, which induces chronic granulomatous infection of the skin and peripheral nerves. The disease ranges from the tuberculoid to the lepromatous forms, depending on the cellular immune response of the host. Chemokines are thought to be involved in the immunopathogenesis of leprosy, but few studies have investigated the expression of chemokine receptors on leukocytes of leprosy patients. In the present study, we evaluated 21 leprosy patients (M/F: 16/5) with a new diagnosis from the Dermatology Outpatient Clinic of the University Hospital, Federal University of Minas Gerais. The control group was composed of 20 healthy members (M/F: 15/5) of the community recruited by means of announcements. The expression of CCR2,...

FISH IMMUNE SYSTEM Gayana
Olabuenaga,Susana E..
The defence against infections is the essential function of the immune system of all Vertebrates. Fish present an efficient and developed immunologic response and in the case of Teleosts, with some similarities to the higher vertebrates. It can be divided into two types: innate or non specific and adquired or specific, both with a humoral and cellular compartments. Their relative importance can change according to the age of fish and is always under the influence of different factors. The most relevant components are mentioned here. There are two factors in fishes which strongly influence the immune response and antibody production, seasonal changes and temperature. The main lymphoid organs are described here: the thymus, the kidney and the spleen, as well...

Hematological profile of beef cattle with divergent residual feed intake, following feed deprivation PAB
Gomes,Rodrigo da Costa; Siqueira,Renata Farinelli de; Ballou,Michael Alan; Stella,Tiago Roberto; Leme,Paulo Roberto.
The objective of this work was to characterize the hematological profiles of steers and bulls, according to residual feed intake (RFI), after feed deprivation. Twenty‑month‑old Nellore steers and bulls were fed feedlot diets for 70 and 56 days, respectively. RFI was calculated as the difference between actual feed intake and expected feed intake. More and less efficient steers and bulls, according to RFI, were subjected to 24 hours of food deprivation. Blood was sampled prior to and following the withdrawal period. Hematological analyses included total and differential white blood cell count, red blood cell count and morphology, and plasma glucose concentration. Variation in RFI did not influence plasma glucose concentration or blood cell profiles. Glucose...

Hepatocytes, rather than leukocytes reverse DNA damage in vivo induced by whole body y-irradiation of mice, as shown by the alkaline comet assay Biol. Res.
PINCHEIRA,JUANA; CARRERA,PILAR; MARCELAIN,KATHERINE; DE LA TORRE,CONSUELO.
DNA damage repair was assessed in quiescent (G0) leukocytes and in hepatocytes of mice, after 1 and 2 hours recovery from a single whole body y-irradiation with 0.5, 1 or 2 Gy. Evaluation of single-strand breaks (SSB) and alkali-labile sites together were carried out by a single-cell electrophoresis at pH>13.0 (alkaline comet assay). In non-irradiated (control) mice, the constitutive, endogenous DNA damage (basal) was around 1.5 times higher in leukocytes than in hepatocytes. Irradiation immediately increased SSB frequency in both cell types, in a dose-dependent manner. Two sequential phases took place during the in vivo repair of the radio-induced DNA lesions. The earliest one, present in both hepatocytes and leukocytes, further increased the SSB...

Immunomodulation of Juvenile Pacu, Piaractus mesopotamicus, by Different β(1-3)(1-6)-D glucan Products BABT
Sabioni,Rafael Estevan; Zanuzzo,Fábio Sabbadin; Cyrino,José Eurico Possebon.
Abstract Stress in intensive fish farming hamper immune function of fish and cause losses by disease outbreaks, a situation that can be minimized, but cannot be completely circumvented, by the use of immunomodulators. Addition of immunomodulators to aquafeeds has thus become a common practice. β-glucan (BG) is one of most studied and effective immunomodulators, aquaculture purposes included. Extracted from cell walls of bacteria, fungi and selected cereals, BG activity depends on the source and extraction methods. This study evaluated effects of two BG products (BG1 and BG2), extracted from Saccharomyces cerevisiae under varying extraction methods and with different immune activity, on the feeding of pacu Piaractus mesopotamicus juveniles. BG1 provided...
